Multiple reasons:
1) Friedrich Wilhelm IV. (the king of Prussia) was uninterested in an imperial crown presented to him by a democratic parliament (a crown from the streets) and coupled with a constitution (even though the Kingdom of Prussia adopted a constitution in 1848/50 as well). He was dedicated to the divine right of kings and only would have accepted an imperial crown that would be modeled after the HRE - meaning that the Free Cities and Princes of the German Confederation would have to present him the imperial authority. And as this necessarily included Austria. Thus it was off the table.
2) The Constitution of the Frankfurt Assembly was far more liberal than the constitution of the Kingdom of Prussia and the later North German Confederation/German Empire of 1871 were nearly totally under Prussian hegemony.
3) The Constitution of the Frankfurt Assembly wasn't accepted by all revolutionaries and further uprisings were a possibility. The fact that Prussia intervened in favor of the german states that suffered from the revolution (like Saxony, Hessen, Baden and Bavaria) did much more to solidify and expand prussian hegemony in Germany.

No. Prussia was the "hope" of some revolutionaries as the Austrian Empire had so many non-german territories that it would be very unlikely that they would become the head of an exclusevly german nation state. And the prussian king and his staff were very capable of levying the early nationalist sentiments in their favor, thus creating the illusion that Prussia was interested in creating a German State - in reality Prussia was interested in creating a (german) state under prussian hegemony; which was eventually achieved in 1871.
