It depends on the joint. IF you just use RTV without a gasket in a joint designed for a gaskt then when you get metal-to-metal contact as you tighten the joint the RTV will be squeezed out to microscopic thickness. Then any relative movement of the joint (mechanical or differential expansion) risks cracking the joint as molecular-thickness RTV has virtually no deformability or compliance left, whereas a gasket can flex and move with the joint (to some degree).

If the joint is designed for RTV only then it will have been designed to avoid stressing the RTV during service; in this case the joint surfaces need to be completely clean to ensure the RTV "wets" both surfaces, or you risk oil, coolant, etc wicking through the joint via capillary action.
